Panagia tou Kastrou (Our Lady of the Castle) was built as a Byzantine Greek Orthodox church around the eleventh century and is the oldest surviving church in the medieval town of Rhodes. Originally a cross-in-square Byzantine church, it was remodelled in the first half of the fourteenth century, credited to Grand Master Helion de Villeneuve, into a three-aisled Gothic basilica with a transept and ribbed cross-vaults. Under the Knights it served as the Roman Catholic cathedral of Rhodes, known as Santa Maria del Castello. Pope John XXII is recorded as having provided funds for the Gothic rebuilding.

After the Ottoman conquest of 1522 the church was converted into a mosque, the Enderun Camii, and remained one until after the Second World War. When the Dodecanese passed to Greece in 1947-1948, the minaret and portico were removed and the building was reconverted into a Christian church. Since 1988 it has housed the Byzantine collection of Rhodes, displaying icons, wall-paintings and medieval sculpture, within the UNESCO World Heritage Medieval City of Rhodes.
